目的观察竹节参总皂苷对冠脉结扎致心力衰竭大鼠的保护作用,并探讨其可能机制。方法采用结扎左冠状动脉前降支法建立大鼠心力衰竭模型,用竹节参总皂苷(50和100mg/kg)灌胃给药,1次/d,连续8W。8W后检测大鼠心功能,然后取血进行血液超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH.Px)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量分析;取心脏,计算大鼠心脏重量指数,分别进行HE、Masson和苦味酸.天狼星红染色并用显微镜、电子显微镜观察,计算心肌梗死面积、心室扩张程度、心肌间质胶原容积分数、观察心肌组织形态学。结果模型组大鼠心功能明显受损,心脏重量指数、心肌梗死面积、心室扩张程度、心肌间质胶原容积分数明显升高(P〈0.05或P〈0.01);血液中SOD、GSH-Px、CAT活性显著下降,MDA含量显著升高;形态学观察发现,心肌细胞变性坏死、胶原纤维大量增生,与假手术组比较具有显著性差异(P〈0.01);用竹节参总皂苷(50和100mg/kg)治疗后,上述异常改变得到了有效改善。结论竹节参总皂苷对冠脉结扎致心力衰竭大鼠的心肌损伤有较好保护作用,其机制可能与清除自由基、抗脂质过氧化,提高机体的抗氧化能力有关。
Objective To investigate the cardioprotective effects of total saponin from Panax japonicus on ligating left anterior de scending branch induced heart failure in rats and its mechanism. Methods Methods: Heart failure model was established in male Wister rats by ligating left anterior descending coronary artery. Therapeutic groups were given total saponin from Panax japonicus (50 and 100 mg/ kg) after ligating left anterior descending branch treatment. After 8 w intervention, heart rate (HR), left ventricular systolic pressure ( LVSP), left ventricular end-diastolic pressure ( LVEDP), maximal rate of the increase of left ventricular pressure ( LV + dp/dtmax), max imal rate of the decrease of left ventricular pressure (LV-dp/dtmin) were examined; SOD, GSH-Px, CAT activities and MDA contents in the blood were assayed, respectively; The myocardial hypertrophy index was investigated ; The myocardial infarct size, degree of ventricular dila tation, myocardial interstitial collagen volume fraction and tissue morphology were investigated by HE, Masson, picric acid - sirius red stai ning and observed by a microscope and an electron microscope. Results In the model, the cardiac function was markedly deteriorated, and the myocardial hypertrophy indexes, the myocardial infarct size, degree of ventricular dilatation, myocardial interstitial collagen volume frac- tion were significantly increased; the activities of SOD GSH-Px and CAT were remarkably decreased, the MDA content was significantly in- creased compared with those of sham-operated group (P 〈 0.01, respectively) ; Morphology observed found myocardial cell degeneration and necrosis, proliferation of collagen fibers. After treated with total saponin from Panax japonicas, the upper abnormal results had been effec tively improved (P 〈 0.05 and P 〈 0.01 respectively).
