目的探讨右归丸治疗肾阳虚证网络化分子调节机制。方法根据肾阳虚证评定量表,选取肾阳虚证患者18名给予右归丸治疗一月,选择疗效最好的8例作治疗前后的基因芯片杂交测试,采用SAM软件筛选显著表达基因并予以基因本体功能注释,采用BiosoGenet工具构建基因功能网络。结果右归丸治疗肾阳虚证的总有效率为87.5%。当假阳性率为5%时获得25个显著表达基因,其中14个基因获得基因本体注释,在分子功能、生物学过程及细胞成分3个方面,分别得到12、14及7个显著性基因本体类。基因功能网络结构可以分为三个模块,模块1(UBC基因群)和模块3(DD1T3基因群)基因功能比较广泛,而模块2(MS4A1基因群)主要为免疫反应功能相关基因。结论本研究从基因功能网络角度,提示调节机体的免疫功能为右归丸治疗肾阳虚证的重要疗效机理之一。
AIM To characterize the networked molecular regulation mechanism of the kidney-yang deficiency treated with Yougui Pills.METHODS Eighteen patients with kidney-yang deficiency were selected for one month treatment with Yougui Pills.Accumulated clinical scores of kidney-yang deficiency were assessed pre-treatment and post-treatment using a 40-factor questionnaire of kidney-yang deficiency.Eight cDNA microarray experiments were performed in eight patients with the best therapeutic efficacy before and after the treatment with Yougui Pills.Significance analysis of microarray(SAM) was performed to procedure to screen differential expressed genes for further gene ontology annotation and network construction.An original analytical approach,BiosoGenet,was applied to constructing a gene network according to their biological interactions of differently expressed genes. RESULTS The total efficiency of Yougui Pills reached 87.5%.Twenty-five significantly expressed genes were detected corresponding to a 5% false discovery rate.The functional analysis of these genes identified fourteen genes annotated with gene ontology categories.There were twelve gene ontology themes enriched in molecular function,fourteen gene ontology themes enriched in cellular components and seven gene ontology themes enriched in biological processes including VBC,DD1T3 and MS4A1 gene batteries.The latter was connected with immunity.CONCLUSION This study indicates that immune function related regulations are main molecular mechanism associated with exact clinical efficiency of Yougui Pills.
