睾酮在雄性生殖中起着重要作用。为探讨1 950 MHz射频电磁辐射对小鼠睾丸间质细胞睾酮分泌功能的影响,采用s Xc辐照系统、GSM talk信号对小鼠睾丸间质细胞进行频率为1 950 MHz、比吸收率(SAR)为3 W/kg的24 h的射频辐射;再采用ELISA方法检测辐照后0、6、24 h及假辐照组细胞上清液中的睾酮含量,并利用Real time-PCR方法测定上述3个时间点及假辐照组细胞中类固醇合成快速调节蛋白(st AR)、细胞色素P450胆固醇侧链裂解酶(P450sc)、雄激素受体(AR)的m RNA的表达水平。结果表明:与假辐照组相比,辐照组细胞上清液中的睾酮含量于照射后6、24 h显著降低;辐照组细胞内的P450scc、AR的m RNA表达水平于照射后0、6、24 h显著降低;而辐照组的st AR m RNA表达水平在辐照后0 h时低于假辐照组,6、24 h时与假辐照组几乎无差异。因此,1 950 MHz射频电磁辐射能通过基因转录水平影响到小鼠睾丸间质细胞st AR、P450scc、AR的m RNA的表达,进而降低睾酮的合成和分泌。
Testosterone plays an important role in male reproduction. To investigate the effect of 1 950 MHz radio-frequency electromagnetic radiation on the testosterone secretion in mice testicular leydig cells, we treated mice testicular leydig cells with radiation of 1 950 MHz and 3 W/kg(SAR) for 24 hours, using an s Xc irradiation system and GSM talk signals. The testosterone contents in the treated cell's supernatants of sham-irradiated group and irradiated group at 0, 6 and 24 hours after continuous irradiation were measured using the ELISA method. The m RNA expression levels of st AR, P450 scc and AR at the above three time points were detected by the real time-PCR method. The results showed that, compared with the sham-irradiated group at the three time points, the irradiated group had testosterone content in supernatants and m RNA expression levels of P450 scc and AR all significantly decreased. However, the st AR m RNA expression level in the irradiation group was lower than that in the sham-irradiated group at 0 h after irradiation, while it was almost the same as that in sham-irradiated group at 6 h and 24 h. It is concluded that 1 950 MHz radio-frequency radiation affects the m RNA expression of st AR, P450 scc and AR in mice leydig cells through the gene transcriptional level, thereby reduces the synthesis and secretion of testosterone.
