目的:通过动物实验探讨中医“髓减致痿”病机的科学性。方法:将60只大鼠按随机数字表法分为空白对照组、假手术组、模型组、阳性对照组、补肾组5组,切除大鼠双侧卵巢制作骨质疏松症模型。阳性对照组灌胃给予戊酸雌二醇,补肾组灌胃给予左归丸,连续给药3个月。采用Micro CT检测股骨BV/TV(%),骨计量学方法检测胫骨骨组织形态。结果:与假手术组比较,模型组大鼠骨丢失率显著增加,股骨BV/TV以及胫骨TBV%均显著减少,而TRS%、TFS%及其TRS/TFS显著增加;同时髓腔中出现大量脂肪细胞,ABS%显著增加,而其他骨髓细胞相对减少。而给予左归丸和戊酸雌二醇后,上述指标在一定程度上发生逆转。结论:切除大鼠卵巢3个月后,在骨量显著丢失的同时,骨髓中脂肪细胞大量增多而其他骨髓细胞明显减少,而补肾能在一定程度上抑制这一情况的发生,证明了中医“髓减致痿”病机的科学性。
Objective: Exploring the scientific nature of Traditional Chinese Medicine etiology of "decrease of marrow leading to bone debility" by animal experiments. Methods: SD female rats were randomly divided into blank control group, sham operation group and ovariectomized model group according to their body weights. After the operation, the model group were randomly divided into OVX group, aquadiol group and Zuogui pill group. For three consecutive months administration, rats were sacrificed and then the tibia and femur were fixed with paraformaldehyde. Longitudinal undecalcified bone sections and toluidine blue staining would be made, and bone tissue morphology and adipocyte volume percentage ( ABS% ) would be measured. The BV/TV( % ) of femur was measured by Micro CT. Results : Compared with the sham operation group, TBV% and BV/TV of ovarieetomized model group were significantly decreased, while its values of ABS% , TRS% , TFS% and TRS/TFS were evidently increased. At the same time, a large number of adipocytes appeared in the bone marrow cavity of OVX group rats, but other marrow cells were relatively reduced. After administration of Zuogui pill or estradiol valerate, the above indexes have been reversed in a certain extent. Conclusion: After three months of ovarian resection in rats, bone mass and other marrow cells of OVX group rats shin were reduced significantly while a large number of adipocytes increased in bone marrow. However, Tonifying Kidney could inhibit the occurrence of above situation in a certain extent. This experiment proved the Traditional Chinese Medicine etiology of " decrease of marrow leading to bone debility" was scientific.
