应用凝集素芯片检测系统性红斑狼疮患者红细胞膜表面糖链表达类型。分别取患者和健康人血液作为实验组和对照组,用低渗液处理红细胞使其破碎,离心制备血影,利用凝集素芯片特异亲和性捕获血影表面糖链,激光扫描仪检测凝集素信号,根据凝集素特异性得到细胞膜表面糖链表达变化。结果表明实验组在凝集素位点ECA、WFA、SBA、UEA-Ⅰ呈现阳性(p〈0.01)且在ConA、PTL-Ⅰ、DBA等凝集素位点信号值上调,在STL、SJA、LCA、GSL-Ⅰ、LEL凝集素位点信号值下调(p〈0.05)。根据凝集素亲和特异性显示,系统性红斑狼疮患者红细胞膜表面Gal、GlcNAc、Fuc等糖链结构表达均增加。为系统性红斑狼疮的研究增添理论依据。
Glycan profiling on the cell membrane of erythrocyte of women with systemic lupus erythematosus(SLE) was detected by lectin array. Blood of women with SLE and normal was taken as treated group and control group respectively, which was disposed by hypotonic solution and centrifugated to obtain the ghost. The ghost was caught by lectins printed on the array according to distinctive glycan-binding specificities. The glycan profiling was scanned by laser fluorescence scanner. The results suggested that treated group was absence of binding signals on ECA, WFA, SBA, UEA- Ⅰ(p〈0.0 1). The signals of treated group were stronger than that of control group on ConA, PTL-Ⅰ, DBA, and weaker on STL, SJA, LCA, GSL-Ⅰ, LEL(p〈0.05). According to the distinctive binding specificities of lectins, Galactose, N-acetylgalactosamine and fucose increased significantly in treatd group. The results provided theory evidence for research of SLE.
