为监测猪场内的蚊蝇是否能携带猪繁殖与呼吸综合征病毒(PRRSV),采集贵州省5个养猪场蚊蝇样本,采用RT-PCR方法引物蚊蝇样本进行PRRSV N基因检测,对检测为阳性样本的PRRSV-NSP2基因进行克隆并测序,最后对NSP2基因进行生物信息学分析。结果显示,从花溪、乌当、毕节3个地方猪场的蚊蝇样本中均扩增出372 bp左右的片段。贵州省蚊蝇样本的3株PRRSV-NSP2基因的核苷酸序列之间存在较高的同源性,可达97.9%~98.3%;蚊蝇样本的PRRSV-NSP2基因片段与贵州猪场猪源PRRSV流行株相比具有较高的核苷酸同源性,高达97.6%~100.0%,与国内最近流行变异毒株的核苷酸同源性为84.3%~98.8%,而与早期国内外分离经典毒株的核苷酸同源性为77.1%~97.7%。结果表明,从贵州省几个猪场内采集的蚊蝇携带PRRSV。
In order to detect porcine reproductive and respiratory syndrome virus(PRRSV) in mosquitoes and flies,the mosquitoes and flies in five pig farms of Guizhu were selected.RT-PCRwas used to clone the PRRSV NSP2 gene,and the sequsences was analyzed.The resultes indicated that the length of the PRRSV N gene fragment in the mosquitoes and flies is 372 bp in three pig farms named Huaxi,Wudang,Bijie.The homology of PRRSV NSP2 gene in the three samples of mosquito and flies is up to 97.9%-98.3%,the homology of PRRSV NSP2 gene segments between samples and Guizhou swine PRRSV strains is up to 97.6%-100.0%,the homology between samples and recently popular variant strains of domestic is up to 84.3%-98.8%,the homology between samples and classical strains isolated is up to 77.1%-97.7%.Experimental results showed that mosquitoes and flies collected from some pig farms in Guizhou carried porcine reproductive and respiratory syndrome virus.
