目的检测良性前列腺增生(BPH)与前列腺癌(PCa)组织标本HER-2及C—mycmRNA相对值,探讨此值对PCa诊断的特异性意义。方法通过实时荧光定量PCR检测63例PCa、37例BPH及3例正常前列腺组织HER-2及C-mycmRNA的表达,比较其在PCa与BPH中组织定量的差异。结果相对于BPH,PCaHER-2及C-mycmRNA表达相对值分别为4.25±0.03,7.24±0.06,PCaHER-2及C-myc表达相对值较BPH高,差异有统计学意义(均为P〈0.05)。结论实时荧光PCR定量检测HER-2及c-mycmRNA为PCa的诊断提供了重要的辅助指标。
Objective To study the HER- 2/C- myc mRNA expression in benign prostatic hyperplasia (BPH) and prostate cancer (PCa) tissue, and to explore the value of HER-2/C-myc mRNA expression in PCa diagnosis. Methods A sensitive, real-time fluorescence quantitative (FQ) PCR assay was used to detect the differential expression of HER-2 and C- myc mRNA in prostate samples from 63 patients with PCa, 37 with BPH and 3 healthy controls. Results The expression levels of HER-2 (4.25+ 0.03) and C-myc (7.24+0.06) in PCa tissue were significantly higher than those in BPH (both P〈0.05). Conclusion The detection of HER-2 and C-myc mRNA expression by FQ-PCR may provide reliable and helpful information for the diagnosis of PCa.
