目的利用γH2AX识别抗体和流式细胞术(FCM),评估职业铅暴露工人外周血淋巴细胞DNA损伤及相关影响因素。方法收集某电瓶车蓄电池厂67名工人和厂外70名对照人群基本资料,应用FCM检测淋巴细胞DNA损伤情况。结果高浓度组、低浓度组、内对照组工人外周血淋巴细胞DNA损伤率和平均荧光强度均高于外对照组,差异有统计学意义(P〈0.05);性别、吸烟、饮酒、工龄对职业铅暴露工人DNA损伤水平差异无统计学意义(P〉0.05);偏相关分析结果显示内对照组DNA损伤率和平均荧光强度与年龄存在相关性(r=-0.430、-0.391,P〈0.05),铅暴露高浓度组平均荧光强度与血铅、δ-ALA存在相关性(r=0.621、-0.697,P〈0.05)。结论职业铅暴露可能致人体外周血淋巴细胞DNA损伤;FCM可作为DNA损伤水平检测的快速简捷方法运用,用于大样本快速的DNA损伤水平监测。
Objective To estimate lymphocytes DNA damage of workers exposed to lead and find related factors.Methods Data was collected from 67 workers of a car battery factory and 70 normal subjects,detecting DNA damage was used by γH2AX recognition antibodies and flow cytometer.Results As compared with control group,the DNA damage rate and fluorescence intensity of workers exposed to lead were significantly higher(P0.05).Gender,smoking,drinking and working years didn,t affect DNA damage level(P0.05).Partial correlation analysis showed that both DNA damage rate and fluorescence intensity had a statistical significant correlation with age,after controlling working years in inner control group(r=-0.430,-0.391,P0.05).We also find significant correlation between fluorescence intensity and blood lead,as well as δ-ALA in high lead exposure group(r=0.621、-0.697,P0.05).Conclusion Occupational lead exposure can cause DNA damage;FCM can be applied as a rapid and easy method to detect DNA damage level for huge samples.
