目的探讨大鼠Th1/Th2细胞平衡与心脏微血管fgl2表达是否存在相关性。方法将雄性大鼠随机分成正常对照组、假手术组和模型组,模型组又分为缺血再灌注3h,5h、24h、3d、7d五个亚组,采用实时荧光定量PCR方法检测各组缺血再灌注区心脏组织fgl2mRNA的表达,ELISA方法检测血清IFN-γ、IL-4水平,以IFN-γ/IL-4比值代表Th1/Th2平衡状况,免疫组化方法检测心脏微血管中fgl2的表达。结果正常对照组与假手术组fgl2表达、Th1/Th2比值均无统计学差异;模型组fg12表达、Th1/Th2比值均显著高于正常对照组和假手术组(P均〈0.05),且于再灌注6h达高峰。免疫组化结果显示,心脏微血管缺血再灌注3h即有fgl2沉积,再灌注6h沉积最明显,之后逐渐减少。Spearman等级相关分析显示,Th1/Th2与fgl2水平呈显著正相关(r=0.867,P〈0.05)。结论Th1/Th2平衡状态与心脏微血管fgl2表达存在相关性。
Objective To explore the relationship between Th1/Th2 imbalance and the microvaseulature fgl2 expression existed in rat heart I/R model. Methods Male SD rats were divided into three groups, control group, sham-operated group and operation-treated group, randomly. The operation-treated group was divided five sabgronps after I/R 3 h, 6 h, 24 h, 3 d, 7 d. The rats were sacrificed at the corresponding time point. Real-time Sybr Green I PCR tested the expression of the microvasculature fgl2-mRNA, serum eytokines IFNγ and IL-4 were tested with the ELISA kits, the ratio of IFN-γ/ IL-4 represented the balance of Th1/Th2. S-P immunohistochemistry assay detected fgl2 expression in the microvasculature. Results There was no significant difference between centrol group and sham-operated group in the expression of microvasculature fgl2, the expression of microvaseulature fgl2 in operation-treated group was significant higher than that in control group and sham-operated group(all P〈0.05) ,the expression of fgl2 attained the peak at the 6 h after I/R. There was no significant difference in Th1/Th2 ratio between the control group and the sham-operated group. The Th1/Th2 ratio of operation-treated group was significant higher than that of control group and sham-operated group( all P 〈 0.05 ) ,the ratio attained the peak at the 6h after I/R. fgl2 could be deposited at the 3 h, and attained the peak at the 6 h after I/R, decreased gradually after the point time. There was a significant positive correlation existing between the ratio of Th1/Th2 and the expression of mierovaseulature fgl2 ( r = 0.867 ,P 〈 0.05). Conclusion The positive relationship existed between Th1/ Th2 imbalance and the microvaseulature fgl2 expression.
