目的动态观察多房棘球绦虫混合重组BCG-EmⅡ/3和BCG-Em14-3-3疫苗免疫小鼠后脾细胞因子的变化。方法将混合疫苗采用鼻腔内接种免疫BALB/c鼠,在免疫后0、2、4、6、8和10周各剖杀4只小鼠,取脾,分离脾细胞,用EmAg或ConA刺激培养,收集脾细胞培养上清液,测定IL-2、IFN-7、TNF-α和IL-4水平。试验设有PBS对照。结果疫苗接种组小鼠脾细胞IL-2、IFN-γ、TNF-α和IL-4分别在免疫后2~10周、4~8周、6~10周和4~6周升高,分别在免疫后6、6、8和4周达最高水平,含量分别为10.0pg/ml、(75.0±6.6)pg/ml、(245.0±83.0)pg/ml和(187.5±16.5)pg/ml。结论多房棘球绦虫混合重组BCG-EmII/3和BCG-Em14—3—3疫苗在免疫早期(2~8周)即可诱导小鼠产生TH1和TH2混合型免疫反应。
Objective To dynamically observe changes of cytokines of splenocytes in mice by immunization with mix recombinant BCG-Em Ⅱ/3 and BCG-Em14-3-3 vaccine of Echinococcus rnultilocularis (Em). Methods BALB/c mice were intranasally vaccinated by the mixed vaccine and killed to get spleen on 0, 2, 4, 6, 8 and 10 weeks post-immunization, respectively, splenocytes were separated to culture by stimulation with EmAg or ConA, their supernatants were gathered to measure level of IL-2, IFN-γ, TNF-α and II.-4 by ELISA Kits, PBS served as control. Results In the groups of immunization, levels of IL-2, IFN-γ, TNF-α and IL-4 increased obviously on 2--10 weeks, 4--8 weeks, 6-10 weeks and 4- 6 weeks, respectively, reached the highest level on 6, 6, 8 and 4 weeks, respectively, their contents were 10.0 pg/ml, (75.0±6.6) pg/ml, (245.0±83.0) pg/ml and (187.5± 16.5) pg/ml, respectively. Conclusion TH1 and TH2 mixed response was induced in mice by mix recombinant BCG-Em Ⅱ/3 and BCG-Eml4-3-3 vaccine on early immunization(2-8 weeks).
