目的:寻找内吗啡肽2(EM2)和μ阿片受体(MOR)在内脏感觉和运动调控过程中发挥作用的形态学证据。方法:(1)麦芽糖凝集素(WGA)示踪与免疫荧光组织化学染色技术相结合的多重标记方法;(2)用稀盐酸刺激胃,制作内脏痛动物模型;(3)免疫荧光组织化学多重染色方法。结果:(1)将WGA注入大鼠胃壁,在结状神经节(NG)内可见WGA标记神经元和WGA/EM2/MOR三标神经元;在孤束核(NTS)可见WGA跨节标记的纤维和终末以及WGA/EM2/MOR三标纤维和终末;在迷走神经运动背核(DMV)可见WGA逆标神经元和WGA/MOR双标神经元及其与EM2阳性纤维或终末形成的紧密接触。(2)将稀盐酸注入胃后,NG和NTS神经元内的FOS表达明显增多;在NG内可见EM2/FOS双标神经元;在NTS内可见EM2阳性纤维和终末与SP受体/FOS双标神经元形成紧密接触。结论:EM2和MOR可能影响内脏感觉和运动功能。
Objective : To look for morphological evidence for endomorphin 2 (EM2) and μ opioid receptor (MOR) exert modulatory effects on the visceral sensation and movement. Methods : ( 1 ) Wheat-germ agglutinin ( WGA ) tracing combined with immunofluorescence histochemical staining; (2) Intragastric perfusion with low concentration of hydrochloric acid was used to make visceral pain model; (3) Immunofluorescence histochemical multi-staining. Results: ( 1 ) After injecting WGA into the stomach wall in the rat, WGA retrogradely labeled neurons and WGA/EM2/MOR triple-labeled neurons were observed in nodose ganglion (NG). WGA transganglionically labeled fibers and terminals and WGA/EM2/ MOR triple-labeled fibers and terminals were observed in solitary tract nucleus (NTS). WGA retrogradely labeled neu- rons and WGA/MOR double-labeled neurons and close contacts between WGA/MOR double-labeled neurons and EM2 immunopositive fibers and terminals were observed in dorsal motor nucleus of the vagus nerve (DMV). (2) After intra- gastric perfusion with hydrochloric acid, FOS protein expression in neurons of the NG and NTS increased markedly ; EM2/ FOS double-labeled neurons were found in the NG; close contacts between substance P receptor (SPR)/FOS double labeled neurons and EM2 immunopositive fibers and terminals were observed in NTS. Conclusion: EM2 and MOR might be involved in modulation of the visceral sensation and movement.
