目的:研究红树林内生真菌Aspergillus sp.085035的次级代谢产物。方法:采用柱色谱和半制备高效液相色谱技术对Aspergillus sp.085035的次级代谢产物进行分离纯化,并利用波谱数据和X-射线单晶衍射等方法鉴定化合物结构;采用MTT法对所得化合物进行细胞毒活性检测。结果:从红树林内生真菌Aspergillus sp.085035的乙酸乙酯粗提物中分离得到9个化合物,分别鉴定为:13R-fumigaclavine C(1)、8'-O-methylasterric acid(2)、1,2-secotrypacidin(3)、rhizoctonic acid(4)、chaetominine(5)、helvolic acid(6)、trypacidin(7)、emodin-8-methylether(8)和fumitremorgin C(9)。结论:化合物1为新天然产物,化合物1、8为首次从曲霉属中分离得到,所有化合物均为首次从该真菌中分离得到,化合物7对MDA-MB-435细胞株具有细胞毒活性。
Objective:To study the secondary metabolites of Aspergillus sp.085035 from South China Sea.Methods:The metabolites of Aspergillus sp.085035 were isolated and purified by silica gel column chromatography and semipreparative HPLC,and their structures were elucidated by MS and NMR spectra,and then confirmed by a single-crystal X-ray crystallographic analysis.The cytotoxicity of the compounds was tested by MTT assay.Results:Nine compounds were isolated from the mangrove endophytic fungus Aspergillus sp.085035,and their structures were identified as 13R-fumigaclavine C (1),8'-O-methylasterric acid (2),1,2-seco-trypacidin (3),rhizoctonic acid (4),chaetominine (5),helvolic acid (6),trypacidin (7),emodin-8-methylether (8) and fumitremorgin C (9).Conclusion:Compound 1 is a new natural product,compounds 1,8 are isolated from Aspergillas sp.genus for the first time,all the compounds are isolated from this furgus for the first time.Compound 7 exhibits cytotoxicity against MDA-MB-435 cells.
