目的探讨回阳生肌方药全方及其拆方对糖尿病皮肤溃疡大鼠创面愈合的影响。方法56只SD大鼠随机分为:正常对照组(对照组)、模型组、回阳生肌方药全方组(全方组)、温阳益气拆方组(温阳组)和活血生肌拆方组(活血组)。采用腹腔注射链脲佐菌素-激素干预-皮肤缺损-塑料环埋置方法制备糖尿病皮肤溃疡动物模型。给予各组相应的凡士林纱条和药膏纱条,计算给药3、7、10、14d创面愈合率,观察创面形态变化,给药14d取各组大鼠创面肉芽组织进行HE染色、Masson染色,观察创面肉芽组织结构改变、胶原分布。结果给药3、7、10、14d,模型组愈合率均低于对照组(P〈0.01);给药3d,仅温阳组愈合率高于模型组、且也高于全方组和活血组(P〈0.05);给药7、10、14d,各给药组愈合率均高于模型组(P〈0.05);给药7、14d,全方组愈合率高于温阳组和活血组(P〈0.05)。给药组肉芽组织生长旺盛,成纤维细胞、毛细血管丰富,炎症细胞较少。结论回阳生肌方药可以有效促进糖尿病皮肤溃疡大鼠创面的愈合,温阳益气拆方在愈合早期作用较明显,活血生肌拆方则在愈合中后期效果较好。
Objective To investigate the influences of Huiyang Shengji Formula and its disassembled formulas on wound healing in rats with diabetic skin ulcer. Methods SD rats (n = 56) were randomly divided into control group, model group, whole formula group (treated with whole Huiyang Shengji Formula) , wenyang group (treated with disassembled formula of warming yang and tonifying qi ) and huoxue group (treated with disassembled formula of activating blood and promoting tissue regeneration). The method of intraperitoneal injection of STZ-hormone intervention-skin defect-embedding of plastic ring was used for establishing model of diabetic skin ulcer. All groups were given petrolatum gauzes or corresponding medicated gauzes, and after 3 d, 7 d, 10 d and 14 d, wound healing rate was calculated and wound morphologic changes were observed. After 14 d, the wound granulation tissue was collected from all groups and given HE staining and Masson staining for observing the structure changes of granulation tissue and collagen distribution. Results The wound healing rate was lower in model group than that in control group (P 〈0.01 ) after 3 d, 7 d, 10 d and 14 d. After 3 d, wound healing rate was higher in wenyang group than that in model group, whole formula group and huoxue group (P 〈 0.05 ). After 7 d, 10 d and 14 d wound healing rate was higher in all medicinal groups than that in model group ( P 〈 0.05 ). After 7 d and 14 d, wound healing rate was higher in whole formula group than that in mod- el group, wenyang group and huoxue group ( P 〈 0.05 ). The granulation tissue grew vigorously, fibro- cytes and capillaries were abundant, and inflammatory cells were less in all medicinal groups. Conclu- sion Huiyang Shengji Formula can effectively improve wound healing in rats with diabetic skin ulcer.
