2012年6月新疆沙湾县某奶牛养殖场爆发疑似牛乳头状肿瘤病,为了确诊该病,本研究采取皮肤病变组织进行病理切片检查;设计牛乳头状瘤病毒简并引物BPVP1/BPVP2对皮肤肿瘤组织样品进行PCR扩增,扩增产物克隆至pMD18-T载体中进行测序、序列分析及遗传进化树构建。结果表明:遗传进化树分析显示新疆沙湾县爆发的牛乳头状肿瘤病是由2型牛乳头瘤病毒所引起,与BPV-1、BPV-13进化关系最近。本研究为新疆地区牛乳头状瘤的科学防控提供了理论依据。
In June 2012,a suspected cutaneous papillomatosis broke out in a dairy farm in Shawan, Xinjiang. In order to diagnose the disease,skin lesions were collected for tissue histopathologieal examination. A pair of primer BPV P1/BPV P2 was designed to detect PBV in skin tumor tissue by PCR technique. The amplified fragment was cloned into pMD18-T vector, and then se- quenced for phylogenetic tree analysis. The result showed that the outbreak of tumor disease is caused by bovine papillomavirus type 2, which was most closely related to BPV-1 and BPV-13. This study was provided a theoretical basis for the scientific pre- vention of bovine papilloma.
