紫外光在临床医学上的众多应用中最值得关注的是紫外光照射自血回输疗法,但其具体机制尚未清楚.基于倒置荧光显微镜光路,在单细胞水平上,分别以连续式和间隔式365 nm长波紫外光辐照射血液中数量最多的白细胞-嗜中性粒细胞,检测其对细胞内钙离子浓度的影响.结果表明,相对低剂量的长波紫外光辐照会使胞内钙浓度升高,而较高剂量的紫外光可导致胞内钙浓度下降,甚至低于起始钙浓度.此外,胞外无钙的情况下,紫外仍可引起胞内钙升高,说明紫外引起胞内钙库释放导致胞内钙浓度升高.为探讨紫外光照射自血回输疗法的作用机理提供一定的实验支持.
As a physical therapy, ultraviolet blood irradiation(UBI) is generally regarded as one of the most successful application in clinics. However, the underlying mechanisms remain largely unclear. In the present work, we investigated the effect of continuous and discontinuous 365 nm ultraviolet A(UVA) on cytosolic calcium concentration([Ca^2+]c) in human neutrophils at the single-cell level based on the inverted fluorescence microscope. The results showed that low-dose UVA irradiation induced [Ca^2 +]c increase, in contrast, high-dose UVA irradiation led to [Ca^2 +]c decrease. In addition, external calcium had no obvious effect on UVA-induced [Ca^2 +]c increase, indicating that calcium store release make a greater contribution to this [Ca^2+]c increase. This work on regulation [Ca^2+]c of neutrophils by UVA might be helpful in the research of UBI.
