目的:探讨DDR1蛋白在氧糖剥夺后的神经元细胞及星形胶质细胞中的表达情况,推测可能机制。方法:采用SD孕鼠的胚鼠皮层培养神经元,采用新生SD大鼠皮层培养星形胶质细胞。建立氧糖剥夺(OGD)损伤模型并随机分为神经元正常对照组、神经元OGD组、星形胶质细胞正常对照组和星形胶质细胞OGD组。噻唑蓝(MTT)比色法测定细胞活性;免疫荧光染色及Westernblot方法检测各组DDR1的表达情况。结果:与正常对照组相比,OGD组细胞形态发生明显改变,MTT法测得细胞活性值降低;与神经元正常对照组相比,神经元OGD组的DDR1蛋白表达水平增加(P<0.05);与星形胶质细胞正常对照组相比,星形胶质细胞OGD组的DDR1蛋白表达水平无明显增加(P>0.05)。结论:OGD处理可诱导DDR1蛋白在大鼠皮层神经元中的表达,提示DDR1蛋白可能参与OGD后细胞凋亡等病理生理过程。
Objective:To investigate the expression of DDR1 in primary culture neurons and astrocytes af-ter OGD ,and to detect its possible mechanism .Methods :Culturing neurons from the cerebral cortex of fetal SD rats and astrocytes from newborn SD rats .Submitted to oxygen glucose deprivation (OGD) and divided randomly into neurons control group ,neurons OGD group ,astrocytes control group and astrocytes OGD group .Cell viability was assayed with M TT method and the expression of DDR1 was determined by immunofluorescence staining and Western blot analysis .Results :The morphology was changed and cell viability was inhibited by OGD .Compare with neurons control group ,the expression of DDR1 was significant increased in neurons OGD group ,however compare with as-trocytes control group ,the expression of DDR1 was not increased in astrocytes OGD group .Conclusion:OGD treat-ment can induce DDR1 protein expression in rat cortex neurons and DDR1 may participate in the pathophysiology process such as apoptosis after OGD .
