首先用二烯酮(1)与甲胺溶液作用制得哌啶酮(2),然后利用Gewald反应将哌啶酮、氰乙酸乙酯和硫磺在吗啡啉存在下进行反应,制得中间体胺(3),再与三苯基膦、六氯乙烷在无水三乙胺存在下,得到中间体膦亚胺(4),接着采用3组分串联aza-Wittig反应方法合成了8个新的2-烷胺基-3-芳基四氢吡啶并噻吩并嘧啶-4(3H)-酮衍生物.所得产物的结构由核磁共振氢谱,质谱及元素分析确证.
In the first, reaction of 1,5-diphenylpenta-1,4-dien-3-one (1) with methylamine yielded 1- methyl-2,6-diphenylpiperidin-4-one (2). Then Gewald reaction was utilized to obtain intermediate amine (3) through the reaction of 2 with ethyl cynoacetate and sulfur in the presence of morpholine. At last, imi- nophosphorane (4) was obtained by the reaction of 3 with triphenylphoshine and hexachloroethane in the presence of triethylamine. Thus eight unreported 2-alkylamino-3-aryl- tetrahydropyrido[3' ,4': 5,4]thieno [2,3-d] pyrimidin-4(3H)-one derivatives (7) was synthesized via tanden aza-Wittig reaction. The struc- ture of the compound (7) was confirmed by 1H NMR, MS and IR analysis.
