烷基间苯二酚(ARs)是小麦麸皮中的酚类类脂,具有多种生理活性,也是粮食行业标准(LS/T 3244—2015)评判全麦粉的重要生物标记物。为高效、稳定地提取全麦粉中的ARs,采用超声波辅助提取手段,研究了超声功率、提取时间及料液比3个因素对ARs提取量的影响。在单因素试验的基础上,通过正交试验对提取工艺参数进行了优化。结果表明:超声波辅助提取全麦粉中ARs的最佳工艺参数为超声功率285 W、提取时间2 min、料液比1∶50(g/mL)。此条件下ARs提取量不低于连续振荡提取法,但将提取时间由48 h缩短至2 min,且样品用量少、重复性好,适用于全麦粉中ARs的快速提取。
Abstract: Alkylresorcinols (ARs), the main phenolic lipids in wheat brans, are of many biological activities, and become the important biomarkers for judgment of whole wheat meal according to the Chinese grain industry standard (LS/T 3244--2015). For the purpose of developing an effective and stable extraction method, the ultrasound technique was applied. The effects of ultrasonic power, extraction time and the ratio of solid to liquid on extraction yield of ARs were investigated in this study. On the basis of single-factor experiment results, the orthogonal experiment was conducted to optimize the conditions for extraction of ARs from whole wheat meal. The optimum parameters were obtained with ultrasonic power of 285 W, extraction time of 2 min, and solid to liquid ratio of 1:50 g/mL, under which the ARs yields by ultrasonic-assisted method were not less than its counterparts by continuous shaking, but the extraction time was greatly shortened from 48 h to 2 min. Therefore, the ultrasonic-assisted method is more suitable for ARs extraction from whole wheat meal with small quantity requirement for sample and good repeatability.
