以自制的琥珀酰化壳聚糖(SUCS)和羟甲基-5氟尿嘧啶(5-FuOH)为原料,制备了壳聚糖偶联载药体(SUCS-5-FuOH)。傅里叶红外光谱(FT—IR)、核磁共振(1H-NMR)、热重(TG)和X粉末衍射(XRD)等技术表征了产物的化学结构、热稳定性和结晶行为。并通过紫外光谱(UV)分析了产物对5-FuOH的接枝率及其体外释药行为。结果表明,SUCS和5-FuOH间通过酯键化学键合,接枝率(载药率)为18.7%,在模拟体液和酶存在条件下均有良好的缓释性能。
Chitosan coupling drug-loading body (SUCS-S-FuOH) was prepared with homemade succinyl chi tosan (SUCS) and hydroxymethyl-5-fluorouracil (5 Fu()H) as materials. The chemical structure, thermal sta- bility and crystallization behavior of the obtained sample were characterized by Fourier infrared spectrum (FT IR), nuclear magnetic resonance ('H NMR), thermal gravimetric analysis (TC-) and X-ray powder diffraction (XRD). The grafting rate of 5 FuOH and the in vitro releasing performance were analysed by UV spectrum (UV). The results indicated that SUCS and 5-FuOH were chemically bonded through ester linkage, the graft ing rate (drug loading rate) of SUCS-5-FuOH was 18.7%, and it had good sustained release performance under the condition of simulated body fluids and enzymes.
