目的探讨转染腺病毒载体对雌二醇促垂体催乳素细胞增殖的影响及可能的分子机制。方法应用BrdU标记法检测转染腺病毒载体后雌二醇促垂体细胞增殖作用的变化,实时荧光定量PCR方法检测腺病毒载体转染组雌激素反应基因ABCG2的mRNAs表达水平,Western Blot方法检测腺病毒载体对垂体前叶细胞磷酸化p38MAPK蛋白表达的影响。结果转染腺病毒载体显著下调了雌二醇的促催乳素细胞增殖作用(P〈0.01),腺病毒转染组磷酸化p38MAPK蛋白表达显著增加,ABCG2基因的表达显著下降(P〈0.01)。结论腺病毒载体转染下调雌二醇促垂体催乳素细胞的增殖作用,其机制可能与腺病毒载体改变p38MAPK和ABCG2的表达有关。
Objective To investigate the effect of adenovirus vectors on the proliferation of pituitary lactotrophs promoted by estrodiol. Methods Effect of adenovirus vectors on the proliferation of pituitary lactotrophs promoted by estrodiol was detected by BrdU labeling. Expression of ABCG2 mRNAs in the lactotrophs transfected by adenovirus vectors was determined by quantitative real-time PCR. Western blot method was used to detect the expression of phospho-p38 MAPK protein in the pituitary cells transfected by adenovirus vectors. Results The effect of lactotrophs proliferation promoted by estrodiol in the adenovirus groups was attenuated significantly compared with the control group (P〈0.01). After the transfection with adenovirus vector, the expression of ABCG2 mRNAs was down-regulated and phosphop38 MAPK protein was up-regulated significantly in the lactotrophs (P〈0.01). Conclusion Adenovirus vectors transfection downregulated the proliferation of pituitary lactotrophs promoted by estrodiol, which might be related to the expression of ABCG2 and p38 MAPK.
