目的探讨异丙酚对哮喘大鼠骨形态构建蛋白4(BMP-4)表达的影响。方法SD大鼠45只,随机分为3组,对照组(Ⅰ组)15只,为健康大鼠;哮喘模型组(Ⅱ组)15只,为卵清白蛋白(OVA)致敏后吸入激发建立模型;异丙酚组(Ⅲ组)15只,为哮喘大鼠给予异丙酚50mg·kg^-1·h^-1,2h。用HE染色后病理图像分析方法检测大鼠气道形态学参数,免疫组化方法检测BMP-4表达定位,Western蛋白印迹法测定BMP-4含量。结果Ⅱ组与Ⅰ组相比,支气管内径缩小,平滑肌层增厚,BMP-4含量减少7。9%(P〈0.05);Ⅲ组与Ⅱ组相比,支气管内径有所增大,肺内含有BMP-4含量增加4.1%(P〈0.05)。结论异丙酚对哮喘大鼠肺内BMP-4表达降低趋势有所缓解,可能是其气道保护作用机制之一。
Objective To investigate the effect of propofol on the expression of BMP-4 in the lung of asthma rat model. Methods Fortyfive SD rats were randomly divided into three groups,15 for each group:group Ⅰ ,healthy rats as control group; group Ⅱ ,asthma group where a asthma rat model was established by sensitization and challenge with ovalbumin (OVA) ; group HI, propofol group, propofol 50 mg· kg^-1· h^-1 was continuously infused iv for 2 hours after the same treatment as group Ⅱ .The change of airways and the expression of BMP-4 were determined by image analysis system,immunohistochemistry and Western blotting. Results BMP-4 was found on the bronchial epithelium,in the vascular endothelium,and on a few scattered alveolar cells of all groups. There were a decline in A1/D and an increase in A2/D in the airways of group Ⅱ as compared with those of group Ⅰ (P 〈 0.05 ). The BMP-4 level in group Ⅱ was lower than that of group Ⅰ (P 〈 0.05). The level of A1/D was higher in group Ⅲ compared with that of group Ⅱ and the level of BMP-4 also increased 4.1%(P 〈 0.05). Conclusion The expression of BMP-4 decreases in the asthma rat model. Propofol attenuates the decrease, which may be one of the mech- anisms of airway protection of propofol.
