为研究鸭坦布苏病毒(DTMUV)E蛋白的免疫原性,用质粒pET32a-E转化Rosseta感受态细胞,经过IPTG诱导表达了重组E蛋白,蛋白大小约为48 ku,Western blot分析表明该蛋白能与DTMUV阳性血清产生特异性反应.将重组E蛋白通过肌肉注射免疫接种28 d麻鸭,采用间接ELISA检测抗体消长情况.结果表明,随着免疫次数的增加及免疫时间的延长,免疫接种麻鸭的抗体水平总体呈逐渐升高趋势.说明融合表达的E蛋白具有良好的免疫原性,能够诱导麻鸭产生有效的免疫应答.
To investigate the immunogenicity of E protein against DTMUV, the recombinant protein E was expressed and detected by Western blot. The molecular weight of E protein is 48 ku. Furthermore, the im- munogenicity was evaluated in 28 d shelducks with the recombinant protein via intramuscular injection. The specific antibodies were detected by indirect ELISA, which showed the antibody titers increased grad- ually in general after multiple E protein inoculations. In conclusion, E protein has good immunogenicity and is able to induce effective immune responses in shelducks.
