目的:分析广西百色地区壮、汉两民族冠状动脉粥样硬化性心脏病(简称冠心病)患者载脂蛋白E基因多态性与冠心病的关系。方法:①分组:冠心病组94例,其中壮族42例,汉族52例,均为右江民族医学院附属医院心内科2005—08/2006—08的住院冠心病患者。全部符合1979年世界卫生组织修订的冠心病诊断标准。对照组100例中壮族50例,汉族50例,均为同期住院的非冠心病患者,并排除心脑血管疾病及肝、肾、内分泌等影响脂质代谢的疾病。②方法:用酚氯仿抽提核酸法从凝血块中分离,用多聚酶链式反应,Hhal内切酶消化法进行载脂蛋白E基因多态性检测。结果:所有受试者完成测试并进入结果分析,无脱落者,DNA抽提均成功。①载脂蛋白E的基因型频率:对照组中壮族人群的E2/2和E2/3基因型频率明显高于汉族人群(P〈0.05),而E3/4基因型频率却明显低于汉族人群(P〈0.05);壮族对照组E2/2和E2/3基因型频率明显高于壮族冠心病组(P〈0.05);而E3/4基因型频率明显低于壮族冠心病组(P〈0.05);汉族对照组E2/3基因型频率明显高于汉族冠心病组(P〈0.05);壮族冠心病组的E2/2基因型频率明显高于汉族冠心病组。②载脂蛋白E等位基因频率:壮族对照组载脂蛋白Eε2等位基因频率明显高于汉族对照组(0.239±0.216,0.101±0.169,P〈0.05),ε3和ε4等位基因频率与对照组比较,差异无显著性(P〉0.05)。壮族和汉族冠心病组ε2等位基因频率明显低于相应对照组(P〈0.05),ε4等位基因频率明显高于相应对照组(P〈0.05)。冠心病组中壮、汉两个民族载脂蛋白E之ε2,ε3和ε4等位基因频率差异无显著性(P〉0.05)。结论:广西壮族自治区百色地区壮、汉两民族冠心病人群中载脂蛋白E基因多态性(ε2等位基因缺失)为冠心病的危险因子
AIM: To analyze association between the polymorphism of apolipopetein E (Apo E) and risk for coenary heart disease (CHD) in Zhuang and Han population fem Beise city of Guangxi Zhuang Autonomous Region. METHODS: (1)Totally 94 patients including 52 Han population and 42 Zhuang population with laboratory verified CHD were fem the Department of Cardiology of Affiliated Hospital of Youjiang Medical College for Nationalities fem August 2005 to August 2006. All subjects met with the CHD diagnostic standard evised by World Health Organization in 1979. 100 contels including 50 Han population and 50 Zhuang population wee at the same time non-coenary heart disease patients without cardiovascular disease, cerebevascular disease and other diseases of liver, kidney, endocnne and so on. (2)DNA was collected by phenoi-chloeform extraction method. DNA was amplified by polymemse chain reaction (PCR) and digested with Hhal to determine Apo E genotypes. RESULTS: All subjects wee involved in the esult analysis, no dep-out and DNA was extracted successfully. (1)Apo E phenotypes: in contel geup, frequency of phenotypes E2/2 and phenotypes E2/3 of Zhuang wee obviously higher than Hart population (P 〈 0.05), and frequency of phenotypes E3/4 was lower (P 〈 0.05). Frequency of phenotypes E2/2 and phenotypes E2/3 of Zhuang population in the contel geup wee higher than in the CHD geup (P 〈 0.05), and frequency of phenotypes E3/4 was lower (P 〈 0.05). Frequency of phenotypes E2/3 of Han population in the contel group was higher than in the CHD group (P 〈 0.05). Frequency of phenotypes E2/2 of Zhuang population was higher than Han population in the CHD group. (2)Apo E allele fequency: In the control group, the fequency of the epsilon (ε) 2 of Zhuang population was markedly higher than Han population (0.239±0.216, 0.101±0.169,P 〈 0.05), and thee was no significant diffeence on the epsilon ε3 and ε4 (P 〉 0.05).
