目的探讨固醇调节元件结合蛋白-1c(SREBP-1c)和c-Jun氨基末端激酶(JNK)在2型糖尿病大鼠肝损伤中的表达和意义。方法高脂饲料喂养结合小剂量链脲菌素建立2型糖尿病伴肝损伤大鼠模型组.同时以普食喂养的大鼠作为对照组,检测两组大鼠血糖、血脂水平,同时检测SREBP-1c及JNK的蛋白表达水平,观察大鼠肝损伤与SREBP-1c及JNK蛋白表达水平之间的关系。结果模型组血糖及血脂水平明显高于对照组,模型组肝功能生化指标检测显示损伤明显,模型组与对照组差异有统计学意义(P〈0.05);模型组SREBP-1c蛋白表达阳性着色区域积分吸光度值为(51767.21±3246.37),JNK阳性表达率为90.9%;对照组阳性着色区域积分吸光度值为(35814.37±2971.52),JNK阳性表达率为31.6%,两组差异有统计学意义(P〈0.05)。结论模型组大鼠SREBP-1c与JNK蛋白表达水平明显高于对照组,2型糖尿病伴肝损伤大鼠的SREBP-1c和JNK蛋白表达明显增加。
Objective To explore the expression and significance of SREBP-1c and JNK in type 2 diabetic rat liver injury. Methods 2 diabetic rats with hepatic injury models were induced by high fat diet with low dose streptozotocin, the rats were fed normal diet as a control group, two groups of blood glucose levels and lipid levels were detected, the protein expression levels of JNK and SREBP-1c were detected. Results There was significant difference of the bio- chemical markers of liver function between the model group and the control group (P 〈 0.05), it was indicated that the protein expression of SREBP-1c and JNK was increased in the liver tissue of model group, there was significant dif- ference of the protein expression of SREBP-1c and JNK between the model group and the control group (P 〈 0.05), Conclusion The model rats group are significantly higher than the control group, SREBP-1c and JNK protein expression are significantly higher in rats with type 2 diabetes joint with liver injury.
