目的研究平糖方对高脂肥胖大鼠糖脂代谢的影响及机制。方法采用高脂喂养大鼠肥胖模型,随机分为高脂模型组、平糖方低剂量(0.42g/kg.d)和高剂量组(0.84g/kg.d),每组10只,另外10只普食大鼠作为正常对照组。给药8周后,采用OGTT和ITT检测胰岛素敏感性,自动分析仪检测血糖血脂,免疫组化H&E染色观察脂肪组织形态,real—timePCR检测脂肪组织AMPK-α1、AMPK-α2、Leptin和TNF-α基因的mRNA表达。结果平糖方干预8周后,大鼠内脏脂肪(肾周和附睾)减少,脂肪细胞体积减小,胰岛素抵抗减轻,TG和LDL降低(P〈0.05),荧光PCR显示平糖方减少脂肪组织AMPK-α2、Leptin和TNF-α1的表达。结论平糖方能减轻高脂大鼠腹型肥胖,改善胰岛素抵抗和糖脂代谢紊乱,其机制与调节脂肪组织AMPK信号和脂肪因子的表达有关。
Objective To investigate the effects and mechanism of Ping-tang recipe (PTF) on Glucose and lipid metabolism in high-fat induced obese rats. Methods Through high-fat diet to build fat rat model, rats in the high-fat diet were randomly allocated into three groups (10 rats each group) : control, high fat diet with treatment of low dose (0. 42g/kg. d) and high dose (0.42g/kg. d)PTF respectively, and another 10 rats were given normal diet as the normal group. After 8 weeks of treatment, Insulin sensitivity were tested by insulin tolerance test (ITT) and oral glucose tolerance test (OGTT), Serum chemistry were analyzed by an automatic chemistry analyzer. Fat tissue morphology were tested by hematoxylin and eosin (H&E) stain, The mRNA change of AMPK, leptin and TNF-α were analyzed by real-time PCR. Results After 8 weeks of PTF treatment, Rats treated with low-dose or high-dose PTF had less epididymal and perirenal adipose tissue compared with the HFD control group(P 〈 0.05 ). fat cell volume decreased and insulin resistance was improved. PTF also decreased serum triacylglycerol (TG) and low density lipoprotein (LDL) level. The expression of AMPK-α2 were increased, and the adipokines including leptin and TNF-α were decreased (P 〈 0.05). Conclusion PTF is effective in reduce visceral fat content, improve insulin resistance, improve glucose and lipid metabolism, the mechanism in part through regulating AMPK signaling and adipokines expression.
