With sugarcane SPS Ⅲ gene alternative splicingintron retention 6,7,8 fragments as the three target sequences,PCR amplification technique prepared three SPSⅢ intron probes labelled with Digoxgenin：SPS-intron 6 probe 167 bp,SPS-intron 7 probe 93 bp and SPS-intron 8 probe 175 bp.Dot-blot hybridization indicated the sensitivity of the probes was high,detecting the pg level of the target nucleic acid,and the specificity was good,for the negative control showed non-hybrid signal.
