目的探讨hsa-miR-26b表达沉默对脂肪细胞蛋白分泌的影响。方法将hsa-miR-26b表达沉默组及对照组慢病毒感染人脂肪前体细胞,在诱导分化过程中收集不同时间点的细胞培养基上清液,采用ELISA法检测IL-6和瘦素(leptin)的分泌量。结果在细胞分化过程中,hsa-miR-26b表达沉默组IL-6与leptin表达量均较对照组降低(P〈0.01或P〈0.05)。结论降低hsa-miR-26b表达在一定程度上可以改善人脂肪细胞炎症状态及胰岛素抵抗,为肥胖治疗提供潜在靶点。
Objective To investigate the effect of hsa-miR-26b-silenced lentivirus on the secretion of adipocytokines during differentiation.Methods Human preadipocytes were infected with hsa-miR-26b-silenced lentivirus(group A)or control lentivirus(group B).The supernatant of culture medium was collected at different time points of differentiation,and the secretions of IL-6and leptin were detected by ELISA.Results Compared with group B,the expressions of IL-6and leptin were decreased during differentiation in group A(P〈0.01or P〈0.05).Conclusion Downregulation of hsa-miR-26bexpression can improve the inflammation and insulin resistance of human adipocytes,which may provide a potential target for the treatment of obesity.
