目的:对五年生人参叶生长发育过程中苹果酸脱氢酶(Malate Dehydrogenase MDH),乳酸脱氢酶(Lactate de-hydrogenase LDH),乙醇脱氢酶(Alcohol dehydrogenase ADH),葡萄糖-6-磷酸脱氢酶(Glucose-6-phosphate Dehydro-genase G6PDH),α-磷酸甘油脱氢酶(α-Glycerophosphate dehydrogenaseα-GPD)5种氧化还原酶的活力进行比较,了解人参叶生长情况。方法:分别于展叶期、开花期、绿果期、红果期、枯萎期采取人参叶样品。采用中性缓冲液提取粗酶液,应用分光光度法测定MDH、LDH、ADH、G6PDH、α-GPD的活力。结果:在人参叶生长发育过程中,MDH、ADH和LDH活力在展叶期明显高于其他时期;LDH、G6PDH在展叶期和结果期末期活力最高;α-GPD在结果期出现活力峰值。结论:通过测量MDH、LDH、ADH、G6PDH、α-GPD的活力变化可判断人参生长情况。
Objective:To compare the activities of Malate Dehydrogenase(MDH),Lactate Dehydrogenase(LDH),Alcohol Dehydrogenase(ADH)and Glucose-6-phosphate Dehydrogenase(G6PDH)and α-Glycerophosphate dehydrogenase(α-GPD)in Ginseng leaves during growth and development,in order to understand the growth of Ginseng leaves.Methods:Take ginseng leaf samples in the spreading stage,the flowering stage,the green fruit stage,the red fruit stage and the withering stage.Adopt neutral buffer solution to extract the enzyme solution of Ginseng leaves.Use spectrophotometry to test the activities of MDH,LDH,ADH,G6PDH and α-GPD.Results:In the growth and development process of Ginseng leaf,the activities of MDH,ADH and LDH in the spreading stage obviously higher than other stages.LDH and G6PDH have the highest activity in the spreading stage and the end of the fruit setting stage,there was a peak of the activity of α-GPD in fruit setting stage.Conclusion:Measuring the activity changes of MDH,LDH,ADH,G6PDH and α-GPD could judge ginseng growth situation.
