目的:探讨下调miR-181a对人胃癌细胞生物学行为的影响。方法:转染miR-181ainhibitor下调miR-181a在人胃癌细胞SGC-7901中的表达。通过MTT法、流式细胞仪、Annexin-V+PI双染色法和Tr-answell实验,观察下调miR-181a表达对人胃癌细胞增殖、凋亡及侵袭迁移等生物学行为的影响。结果:瞬时转染miR-181ainhibitor后胃癌细胞SGC-7901中miR-181a的表达明显下调(P=0.0027)。转染miR-181ainhibitor后7901细胞增殖明显减弱(P=0.0006),被阻滞的G0/G1期细胞明显增多(P=0.0075),S期细胞明显减少(P=0.0022),细胞凋亡比例增加(P〈0.0017)。Transwell实验证实下调miR-181a表达能促进人胃癌细胞侵袭与迁移(P=0.0006、P=0.0004)。结论:下调miR-181a表达抑制胃癌细胞增殖、促进凋亡,促进侵袭迁移,提示miR-181在胃癌的发生发展中可能发挥重要的作用。
Objective:To investigate the influence of down -regulating miR - 181a on biological behavior of gastric cancer cell. Methods: miR- 181a inhibitor was transfected transiently into gastric cancer cell SGC -7901 to down - regulate the expression of miR - 181a. The influence of down - regulation of miR - 181a on cell proliferation, apoptosis,invasion and migration were evaluated by MTT,flow cytometry,Annexin -V + PI and transwell assay. Results: The transfection of miR- 181a inhibitor down- regulated miR- 181a expression in SGC -7901 (P = 0. 002 7). In SGC -7901 cell after transfection with miR- 181a inhibitor, the proliferative ability was inhibited (P = 0. 000 6), G0/G1 phase cell ratio was arrested (P = 0. 007 5), S phase cell ratio was decreased (P = 0. 002 2), and apoptosis rate was increased (P 〈 0. 001 7). Transwell assay showed cell invasion and migration was increased greatly in miR - 181 a down - regulated SGC - 7901 cell ( P = 0. 000 6, P = 0. 000 4 ). Conclusion : Down - regulation of miR - 181 a inhibits gastric cancer cell proliferation and promotes apoptosis, but enhances cell invasion and migration. It may play an important role in gastric carcinogenesis.
