小脑顶核刺激对急性心肌梗死大鼠心脏神经功能的影响

目的:观察小脑顶核刺激(FNS)对急性心肌梗死(AMI)大鼠自主神经功能的影响。方法:成年雄性Sprague-Dawley(SD)大鼠100只,分为:①AMI组(30只),单纯结扎大鼠左冠状动脉(LCA);②FNS组(30只):结扎大鼠LCA,1h后电刺激左侧小脑顶核1h;③FNL组(30只),鹅膏氨酸毁损两侧小脑顶核,5d后再按②方法操作;④Sham组(10只),即假手术组,开胸手术后仅在LCA下穿线但不结扎,电极插入小脑顶核,不予刺激。各组在开胸术后6h、24h、7d、21d4个时间点,进行心率变异性(HRV)的频域分析。21d记录心脏血流动力学参数并计算心肌梗死面积。结果:结扎6h后FNS组频域分析指标-低频(Low Frequency,LF)低于AMI和FNL组(P〈0.01),同时FNS组高频(High Frequency,HF)明显高于其他干预组(P〈0.01)。结扎1d后,FNS组仍然呈现HF成分的增加和LF成分的下降(P〈0.05)。7d后只有假手术组保留了较高的HF和较低的LF,各干预组之间的差别消失。21d后各组之间HRV参数无差别。FNL组与AMI组比较,各时间点HRV参数差异无统计学意义(P〉0.05)。结扎后21d比较各组血流动力学指标,心肌梗死面积也无差别(P〉0.05)。结论:FNS可改善AMI后大鼠心脏神经功能。

英文摘要:

Objective:To observe the effect of fastigial nucleus stimulation (FNS) on autonomic nerve functions of surgically induced acute myocardial infarction rats. Method: 100 Sprague-Dawley rats were randomly allocated in four groups, including AMI group with coronary arteries ligated, FNS group with coronary arteries ligated and fastigial nucleus stimulated one hour later, FNL group with fastigial nucleus lesioned before, then coronary arteries ligated and fastigial nucleus stimulated, and sham-operation group. The characteristics of heart rate variability were determined 6 h , 1 d , 7 d and 21 d after the ligation, and hemodynamic parameters and infarction sizes were compare 21 d. Result:Compared with AMI and FNL groups, FNS group showed lower LF and high HF at 6 h and 1 d. After 1 d only sham operation group remained high HF and low LF. There was no differences in heart rate variability, hemodynamic parameters and infarction size at 21 d between groups. Conclusion.. FNS may has protective effects on cadiac autonomous nerve functions of surgically induced myocardial infarction rats.
