目的:探讨不同补肾方法对化疗导致的卵巢早衰(POF)大鼠卵巢功能的影响.方法:以连续腹腔注射环磷酰胺(CTX)14 d的方法建立卵巢早衰模型.之后分别连续灌服补肾阴中药(六味地黄丸)和补肾阳中药(金匮肾气丸)复方28 d,以青年大鼠为空白对照,观察卵巢的形态学变化,并计数各级卵泡及黄体数目,酶联免疫法检测外周血雌二醇(E2)、卵泡刺激素(FSH)和抗卵巢抗体(AoAb)水平.结果:POF大鼠卵巢内各级卵泡及黄体数目减少,外周血E2下降,FSH 升高,AoAb升高.补肾方药能使各级卵泡及黄体数增加,提高体内E2水平,降低血FSH水平,抑制AoAb产生,尤以补肾阴组作用显著.结论:补肾方药能减轻化疗导致的卵巢功能损害,尤其以补肾阴中药效果更显著.
AIM: To compare the different effects of some prescriptions for reinforcing the kidney in different way on ovarian function of mice with chemotherapy - induced premature ovarian failure (POF). METHODS : The model of POF in female mice was established by intraperitoneal injection of eyclophosphamide (cytoxan, CTX) for 14 d constant- ly. The mice were treated for the prescriptions for reinforcing the kidney -Yin (Liuwei Dihuang Wan, LDW) and that for reinforcing the kidney- Yang (Jingni Shenqi Wan, JSW) for 28 d. The young mice without treatment were used as the blank control. The morphological changes of the ovary were observed and the number of follicles in every stage and corpora lutea were counted. The levels of estradiol ( E2 ), follicle - stimulating hormone (FSH) and antiovarian antibody (AoAb) in peripheral blood were determined by ELISA. RESULTS: The number of follicles in every stage and corpora lutea de- creased in the ovaries of POF mice. The level of E2 decreased, the level of FSH increased, and AoAb was positive in pe- ripheral blood of POF mice. The prescriptions for reinforcing the kidney increased the number of follicles in every stage and corpora lutea, increased E2 level, reduced the increasing FSH level and inhibited the production of AoAb. The efficacy was more significant in the group of prescription for reinforcing the kidney -Yin (LDW) than that in the group of prescription for reinforcing the kidney - Yang (JSW). CONCLUSION: The prescriptions for reinforcing the kidney can relieve the chemotherapy - induced ovary injury especially in the group of reinforcing the kidney - Yin.
