复方菖蒲益智汤对脑缺血再灌注损伤小鼠脑组织谷氨酸及血清CaN活性影响的研究

目的观察复方菖蒲益智汤对脑缺血再灌注损伤小鼠急性期脑组织谷氨酸及血清钙调神经磷酸酶(CaN)活性的影响。方法将实验小鼠随机分为假手术组、模型组、复方菖蒲益智汤高剂量组(高剂量组)、复方菖蒲益智汤低剂量组(低剂量组)、尼莫地平对照组(尼莫地平组),每组10只。采用双侧颈总动脉夹闭的方法制备脑缺血再灌注损伤小鼠模型,给药14天后,观察海马区脑组织的病理形态学变化,测定脑组织谷氨酸及血清CaN(钙调神经磷酸酶)活性。结果各用药组小鼠海马区锥体细胞病变较模型组均有所改善,高剂量组改善最为显著;高剂量组小鼠脑组织谷氨酸活性、血清CaN活性均低于模型组、低剂量组及尼莫地平组(P〈0.05)。结论复方菖蒲益智汤能够降低脑缺血再灌注小鼠脑组织谷氨酸、血清CaN活性,对于小鼠脑缺血再灌注损伤有保护作用。

英文摘要:

Objective To observe the effects of Compound Changpu Yizhi Decoction on the the activity of Glutamate( Glu) in the brain tissue and serum Calcineurin-phosphatase( CaN) of model mice at the acute stage of cerebral ischemia and reperfusion injury( CIRI). Methods 50 mice were randomly divided into five groups: shame operation group,model group,high dosage of Compound Changpu Yizhi Decoction group( high dosage group),low dosage of Compound Changpu Yizhi Decoction group( low dosage group),Nimodipine control group with 10 in each group. The model mice with CIRI were established through ligation on both sides of carotid arteries except sham-operation group. After 14 days,the pathomorphological changes of cerebral tissue in Hippocampus were observed,the activity of Glu in the brain tissue and serum CaN measured. Results The cone cell lesion in Hippocampus in mice of medicinal groups had been improved more than that in the model group,especially more significant in the high dosage group; the activity of Glu in the brain tissue and Calcineurin phosphatase( CaN) in the serum in high dosage group was more decreased than that of the model group,low dosage group and the Nimodipine group( P〈0. 05). Conclusion Compound Changpu Yizhi Decoction may reduce the activity of Glu in the brain tissue and Calcineurin phosphatase( CaN) in the serum of mice with CIRI and has a protective action on the condition.
