以河蚬(Corbicula fluminea)为材料,经过甲醇抽提。乙酸乙酯萃取等方法得到河蚬提取物(CFE).用MTT法检测其对肝癌细胞株的抑制作用,借助光学倒置显微镜以及流式细胞术考察CFE对细胞的形态及细胞周期的影响.实验结果表明:CFE对肝癌细胞SMMC-7721有增殖抑制作用,处理72h细胞形态发生了明显变化。其IG50值为70μg/mL.FMC的结果显示,经CFE处理后的肝癌SMMC-7721细胞出现明显的凋亡峰.这说明CFE有效抑制了体外培养的肝癌细胞增殖。并且能够改变癌细胞的形态及细胞周期的分布,诱导癌细胞出现凋亡.
We obtained ethyl acetate extract (CFE) of Corbicula fluminea, and then investigated the antitumor activity of CFE. The inhibition rate of cancer cell was measured by MTT method. The morphological change of SMMC-7721 ceils was detected by inverted microscopy. The effects of CFE on cell cycles of SMMC-7721 were observed by FCM. The results showed that CFE displayed strong growth inhibitory effect in a dose and time dependant manner againsted hepatoearcinoma SMMC-7721 ceils with estimated ICso values of 70 μg/mL after 72 h of treatment with CFE. The morphological change of SMMC-7721 was significant. In the mean time,flow cytometry analysis showed that CFE could induce the emergence of the peak of apoptosis. These results revealed that CFE inhibited the proliferation and induced the apoptosis of the human hepatocarcinoma SMMC-7721 cells in vitro.
