目的:探讨生理、病理状态下大鼠上肢手掌不同区域血含量和上肢手掌皮肤黑色素变化,为气色形态手诊乃至中医学的“司外揣内”提供合理的研究方法和实验依据。方法随机将SD雄性大鼠20只分为正常组和模型组,每组各10只。利用无水乙醇灌胃复制急性胃损伤模型后,两组分别从大鼠尾静脉中给予一定浓度的伊文斯兰(EB)生理盐溶液。通过测量手掌不同部位组织内EB的吸光度(A)值大小,间接反映手掌各区域血含量的变化情况。利用多巴氧化酶检测两组大鼠前爪皮肤黑色素变化。结果①模型组大鼠前爪5区的A 值(0.96±0.46)显著高于正常组(0.35±0.17),差异有高度统计学意义(P〈0.01)。②模型组大鼠5区的黑色素细胞增多。结论应激性胃溃疡的大鼠的病理变化可以在手掌特定区域反映出,初步推断这种变化是通过血流量的变化和手掌皮肤的黑色素变化实现的。
Objective To investigate the change of blood content and melanin in different area of forepaw skin in rat under physiological and pathological conditions, in order to provide reasonable research methods and experimental basis for hand diagnosis and Qisexingtai of traditional Chinese medicine. Methods Twenty SD rats were randomly divided into two groups, normal group and model group, with 20 rats in each group. Acute gastric mucosal injury model was established by intragastric administration of anhydrou ethanol. Rats tail vesns were injected a certain concentration Evan blue (EB) saline solution. Then, by detecting the photo-absorption rate (A value) of the EB in the tissues of the different zones of forepaws, the changes of blood contents in the different areas was examine indirectly. The melanin of rat forepaw skin was detected by dopa oxidase. Resluts ①The A values in 5 zones of model group (0.96±0.46) was higher than that of normal group (0.35±0.12), the differences was statistically significant (P〈0.05).②The melanin in 5 zones were significantly increased. Conclusion The pathological changes of stress ulcer in rats can be reflected on particular area of forepaw in acute gastric mucosa injury rats, preliminary conclusion the change is realized through the blood flow change and the palmar skin melanin change.
