目的观察补肾填精方左归丸对快速老化模型小鼠骨组织破骨细胞功能的影响,探讨其作用机制是否与干预活化的T细胞转录核因子蛋白表达有关。方法取SAMR1同源对照组小鼠8只为对照组(SAMR1),取SAMP6衰老模型小鼠16只,随机分为模型组(SAMP6)、左归丸组(ZGP),每组8只。灌胃给药12周后收集各组小鼠股骨组织,采用HE染色法观察股骨组织形态,采用TRAP染色法观察股骨组织破骨细胞数,采用Western-blot法检测股骨组织NFAT2蛋白表达水平。结果与SAMR1比较,SAMP6股骨组织形态明显改变、破骨细胞数增多、NFAT2蛋白表达显著升高;与SAMP6比较,补肾填精方能够部分改善模型小鼠的股骨组织形态、减少破骨细胞数、下调NFAT2蛋白表达。结论补肾填精方左归丸对快速老化模型小鼠骨质疏松具有一定的防治作用,其作用机制可能与其通过干预NFAT2蛋白表达调节破骨细胞功能有关。
Objective To observe the effect of Zuogui Pill on osteoclast function in SAMP6 mice,and to research the possibility mechanisms of Zuogui Pill on osteoclast function by regulating the protein expression of NFAT2. Methods The mice were randomly divided into 3 groups,including Senescence Resistant Inbred Strains( SAMR1) group,Senescence Accelerated Mouse Prone 6( SAMP6) group and Zuogui Pill( ZGP) group,8 mice in each group. After 12 weeks' gavage administration,the bone tissue of mice were collected. The morphology of femur was observed through HE staining and the number of osteoclast was observed through TRAP staining. The expression level of NFAT2 in bone tissue was examined through Western blot method. Results Compared with SAMP6 group,Bu Shen Tian Jing recipe can improve the histology morphology,decrease the number of osteoclasts and down-regulation the expression level of NFAT2 of femur. Conclusion Bu Shen Tian Jian recipe can prevent and treat osteoporosis,and the possibility mechanism may be related to Zuogui Pill on osteoclast function by regulating the protein expression of NFAT2.
