目的:研究妊娠恒河猴黄体中甾类激素灵敏调节蛋白(StAR)、转移生长因子(TGF-β1)及其Ⅱ型受体(TβR-Ⅱ)在妊娠早期恒河猴黄体中的表达和IFN-γ的调节作用。方法:利用原位杂交和免疫组化检测mRNA及其蛋白表达与定位。结果:StAR,TGF-β1及TβR—Ⅱ在妊娠恒河猴黄体中的表达随其功能从第15天到第35天逐渐降低;IFN-γ抑制TGF-β1、TβR-Ⅱ和StAR的表达。结论:TGF-β1及其受体对黄体的形成和功能维持可能起重要作用;IFN-γ下调StAR、TGF-β1及其受体的表达。
AIM: To examine expression of transforming growth factor-beta 1(TGF-β1) and TGFβ receptor Ⅱ (TβR-Ⅱ) and steroidogenic acute regulatory protein (STAR) in the corpus luteum (CL) of pregnant monkeys at various stages and to study possible effect of IFN-γon their production. METHODS: In situ hybridization and immunohistochemistry were applied to detect mRNA and protein. RESULTS: The expression of STAR, TGF-β1, and TβR-Ⅱ in the pregnant monkey CL was progressively decreased from d 15 to d 35 of gestation. IFN-γ down-regulated the expression of TGF-β1, TβR-Ⅱ, as well as STAR. CONCLUSION: TGF-β1 may play an important role in the CL formation and functional maintaining; IFN-γ down-regulates the expression of TGF-β1, TβR-Ⅱ, and StAR.
