目的:研究辅助性T细胞亚群(Th17、Th1和Th2)在肺纤维化模型小鼠的动态表达。方法40只雄性C57/BL6小鼠随机均分为两组,分别经气管一次性注入生理盐水(A组)或博来霉素溶液0.04ml(5mg/kg,B组),于第3、7、14和28天处死。采用HE和Masson染色评价小鼠肺组织病理形态变化;检测肺组织中羟脯氨酸(HYP)含量;应用流式细胞术分析脾脏中Th1、Th2和Th17细胞的动态变化。结果A组小鼠肺泡形态正常,肺组织HYP含量无明显变化。B组小鼠第7天时即有部分肺泡结构破坏,肺组织可见大量炎性细胞及红细胞浸润,伴细束状胶原纤维增生;第7、14、28天肺组织HYP含量高于A组(P<0.05);与A组比较,B组4个时间点的Th17和Th2细胞数增加,Th1细胞数减少(P<0.05)。结论Th17、Th2和Th1细胞与特发性肺纤维化密切相关,可能参与其发病机制。

英文摘要:

Objective To investigate the dynamic expression of T helper (Th) subgroups (Th17 ,Th1 and Th2 cells) in mice model with bleomycin‐induced pulmonary fibrosis .Methods Forty C57/BL6 mice were randomly divided into 2 groups with 20 mice each .The mice in group B were intratracheally injected with bleomycin 5 mg/kg to induce pulmonary fibrosis and those in group A used normal saline as the controls .The mice were sacrificed respectively on the 3rd ,7th ,14th and 28th day after administration for observing pathological changes of the lungs(by HE and Masson staining) , determining the contents of hydroxyproline in lung tissues ,and detecting the proportions of Th1 ,Th2 and Th17 cells in the spleen(by flow cytometry) .Results There were no significant changes in lung pathology ,the contents of hydroxyproline in lung tissues ,and the proportions of Th1 ,Th2 and Th17 cells in the spleen during 4 time points in group A .A portion of the alveolar structure was destroyed with a large number of inflammatory cells and red blood cells infiltration and wispy bouquet of collagen fiber hyperplasia started on the 7th day in group B .The contents of hydroxyproline in lung tissues on the 7th ,14th and 28th day were higher in group B than those in group A (P〈0 .05) .The proportions of Th17 and Th2 cells were higher ,but those of Th1 cells were lower in group B than those in group A at all 4 time points(P〈0 .05) .Conclusion Th17 ,Th1 and Th2 cells are closely associated with idiopathic pulmonary fibrosis (IPF) ,which may be involved in the pathogenesis of IPF .
