目的观察芍药苷(抗炎类中药活性成分)对肿瘤坏死因子-α(TNF-α)诱导的人成纤滑膜细胞(FLS)功能的影响。方法体外培养人FLS,考察TNF-α浓度在0.02—20μg·L^-1时对FLS的增殖反应和FLS分泌前列腺素E2、环磷腺苷和白介素-1β的影响;加入芍药苷(1×10^-8~1×10^-4mol·L^-1)后,观察对其增殖反应和分泌功能的影响;用MTT法检测FLS的增殖反应,用放射免疫测定法检测白介素-1β,用酶联免疫吸附法检测前列腺素E2和环磷腺苷的水平。结果TNF-α浓度在0.02~20.00μg·L^-1时,均显著抑制FLS分泌环磷腺苷,显著促进前列腺素E2和白介素-1β的分泌;芍药苷能显著抑制TNF—α对FLS的促增殖反应,明显上调降低的环磷腺苷水平,明显下凋升高的前列腺素E2和白介素-1β水平,且具有一定的浓度依赖性。结论TNF-α可以促进FLS的增殖,降低环磷腺苷,升高前列腺素E2和白介素-1β;芍药苷则能明显逆转TNF-α对FLS的影响。
Objective To investigate effects of paeoniflorin (Pae) on functions of fibroblast - like synoviocytes (FI,S) of human under the stimulation of tumor necrosis factor - α(TNF - α). Methods FLS of human were cultured in vitro. The TNF - α of 0. 02 - 20. 00 μg · L^-1 was incubated with the FLS to measure the proliferative rate of FLS by a MTF assay and levels of interleukin - 1β ( IL - 1β ) by radioimmunoassay;prostaglandins E2 (PGE2) and cyclic adenosine monophosphate (cAMP)by ELISA. The added - on effect of Pae(1 × 10^-8 ~ 1 × 10^-4 mol · L^- 1 ) on the proliferative human of FLS and levels of PGE2 , cAMP and IL - 1β were determined at the concentration of TNF - α (20 μg · L^-1 ) . Results TNF - α ( 0. 02 ~ 20.00 μg · L ^- 1 ) significantly all stimulated the proliferation of the cultured FLS, reducing level of the cAMP and increasing the levels of PGE2 and IL - 1β. Pae suppressed the TNF- α stimulation on FLS, manifesting the up-regulation of the reduced cAMP level and down -regulation of the increased PGE2 and IL- 1β levels in a concentration - dependent manner. Conclusion Pae showed a capacity to reverse the TNF - α stimulation on the FLS of human.
