目的客观量化鹌鹑尿酸(UA)、粪尿及舌象等指标,探讨高尿酸血症鹌鹑模型的中医证候分型,为鹌鹑证候模型评价研究提供参考。方法采用高嘌呤饮食诱导鹌鹁高尿酸血症模型,动态观察鹌鹑尿酸水平、粪尿性状及舌象并进行数字化定量分析。运用健脾利湿中药菊苣进行反证,从“四诊”及以药测证两个角度探讨高尿酸血症鹌鹑中医证候类型。结果问诊、闻诊及望诊显示,实验第17、30天模型组鹌鹑粪尿湿重有升高趋势,粪尿较溏稀气味较大,正常组鹌鹑舌色以红色调为主,模型组鹌鹑舌色变淡呈淡白舌;第22、32天舌象红色度(redintensity,r)及红色面积比例显著下降;与正常组比,第14、21天血清尿酸水平显著升高(P〈0.05)。与模型组比,菊苣组粪尿湿重、干重显著升高(P〈0.05);r及红色面积比例显著上升(P〈0.05);第21天菊苣组尿酸水平显著降低(P〈0.05)。结论采用高嘌呤饮食诱导法,可成功诱导鹌鹑高尿酸血症。量化“四诊”指标及以药测证研究提示:鹌鹑模型部分病理改变与饮食不节导致的“脾虚痰湿”证候有一定的相似之处。
Objective To quantify objectively the indexes of uric acid (UA), excrement, urine and tongue manifestation in quail model of hyperuricemia, investigate the types of TCM syndromes, and provide reference for reviewing quail model of TCM syndromes. Methods The quail model of hyperuricemia was induced by high purine diet. The changes of UA level, characters of excrement and urine and tongue manifestation were observed dynamically, which was then given disproof with Juju (Herbacichorii) with the actions of fortifying spleen and draining dampness. The types of TCM syndromes were discussed in the quail model from two angles including four examinations and inferring syndromes by medicinal. Results The results of inquiry, listening-smelling and inspection showed that the net weight of excrement and urine trended to increase and dilute with strong smell in model group on the 17ts day and 30th day. The color of tongue body is mainly red in normal group and pale in model group. The proportion of tongue redness r and red area decreased significantly on the 22nd day and 32"d day, and UA level increased significantly on the 14'h day and 21st day in model group(P 〈0.05 ). The net weight and dry weight of excrement and urine ( P 〈 O. 05 ) , and proportion of tongue redness r and red area increased significantly in Juju group ( P 〈 0.05 ) compared with model group. The level of UA decreased significantly in Juju group on the 21st day (P 〈 0.05 ). Conclusion The quail model of hyperuricemia can be successfully induced by high purine diet. The quantified study on indexes of four examinations and inferring syndromes by medicinal indicates that part pathological changes of quail model is similar to spleen deficiency and phlegm-dampness syndrome induced by disorder diet.
