目的:探讨P19细胞向心肌细胞分化前后p22-phox水平的变化。方法贴壁培养P19细胞,取合适细胞株,应用0.9%二甲基亚砜( DMSO)诱导培养,收集诱导不同时间的细胞,通过Western blot检测其肌钙蛋白I( cTnI)水平,以确定分化,并行Western blot检测P19细胞向心肌细胞分化前后细胞中 p22-phox 蛋白水平。结果① P19细胞经0.9% DMSO诱导分化后,于第8天开始检测到cTnI表达阳性,后cTnI 表达增加,差异有统计学意义( P 〈0.05)。②P19细胞向心肌细胞分化后细胞内p22-phox水平较分化前高,差异有统计学意义( P〈0.05)。结论 p22-phox在P19细胞向心肌细胞分化后表达增加,氧化应激水平增高。
Objective To investigate the change of the expression of p22-phox before and after P19 cells differenti-ating to cardiac myocytes. Methods Appropriate P19 cells were chosen which were cultured in vitro with 0. 9%dimethyl sulfoxide( DMSO) , then the induced cells were collected at different times. Western blot of cardiac tropo-nin I ( cTnI) was used to identify cell differentiation, and detect the level of p22-phox before and after the differen-tiation. Results ① On the 8th day cTnI was detected positive in P19 cells which were cultured with 0. 9% DM-SO, then the expression of cTnI was increased, with statistical significance(P〈0. 05).② The expression of p22-phox in differentiated P19 cells was higher than that in undifferentiated P19 cells, with statistical significance( P〈0. 05) . Conclusion The expression of p22-phox increases during the differentiation of P19 cells to cardiac myo-cytes, and the level of oxidative stress increases.
