目的:通过网络药理学方法,探讨黄连解毒汤治疗肝炎和肝纤维化的作用机制。方法:结合TDT数据库和PharmMapper Server数据库,筛选出黄连解毒汤的作用靶点;利用OMIM数据库,分别筛选出肝炎和肝纤维化的疾病作用靶点;将以上数据导入PPI数据库进行综合分析,进一步得出黄连解毒汤治疗肝炎和肝纤维化的"化学成分-靶点-疾病"交互网络,再将该交互网络Xgmml文件导入Cytoscapy 3.0软件进行网络可视化和各节点的拓扑参数分析,得出黄连解毒汤抗肝炎和肝纤维化相应的作用机制。结果:黄连解毒汤对肝炎的治疗作用可能是通过干预机体半乳糖凝集素3、表皮生长因子受体、磷脂酰肌醇3-激酶p85α亚基相关信号通路来实现的;而其治疗肝纤维化的作用可能是通过干预半乳糖凝集素3相关信号通路来实现的。结论:黄连解毒汤可以通过干预机体半乳糖凝集素3相关通路达到治疗肝炎的目的,并阻止肝炎向肝纤维化发展。网络药理学方法可作为药物治疗特定疾病的生物机制的计算机预测方法,对药物的发现和研究具有很好的辅助作用。
OBJECTIVE: To probe into the mechanism of therapeutic action of Huanglianjiedu decoction in treatment of hepatitis and liver fibrosis through network pharmacology. METHODS: To select the targets of Huanglianjiedu decoction through TDT database and Pharm Mapper Server data base; and to select the targets of hepatitis and liver fibrosis through OMIM database. The targets of the data were analyzed with PPI database,and then the interactive network of "compound-target-disease " could be obtained; in addition,and the interactive network Xgmml was analyzed with Cytoscapy 3. 0 software for network visualization and the topology parameter value of the dots,and the mechanism of therapeutic action of Huanglianjiedu decoction was obtained. RESULTS: The therapeutic effects of Huanglianjiedu decoction in treatment of hepatitis might concentrate on intervening the related signaling pathways of the Galectin-3,epidermal growth factor receptor,PI3-kinase subunit p85-alpha; while the liver fibrosis might concentrate on intervening the related signaling pathways of the Galectin-3. CONCLUSIONS: Huanglianjiedu decoction can in treatment of hepatitis through intervening the related signaling pathways of the Galectin-3,and prevent the development of hepatitis to liver fibrosis. Meanwhile,network pharmacology method can be regarded as the prediction method of computer in biological mechanism of medication treatment for the specific diseases,which can be regarded as a good method for the discovery and research of medicine.
