目的探讨益肾活血胶囊对同型半胱氨酸(Hcy)所致兔动脉粥样硬化(AS)血管细胞黏附分子-1(VCAM-1)基因与蛋白表达的影响。方法40只新西兰兔,随机分为正常组、模型组、益肾活血胶囊小剂量组、益肾活血胶囊大剂量组及西药(叶酸、维生素B6、维生素B12)组,除正常组外,其他组建立高Hcy血症及AS动物模型,然后分别给予相应的药物治疗。高效液相色谱技术检测血浆Hcy浓度,HE染色观察血管形态学变化,实时定量PCR检测动脉壁VCAM-1基因表达,Western印迹法检测VCAM-1蛋白表达。结果益肾活血胶囊和叶酸等维生素均可明显降低血浆Hcy水平,抑制血管内膜增厚,下调腹主动脉VCAM-1基因和蛋白表达。益肾活血胶囊大剂量组疗效明显优于小剂量组和西药组。结论益肾活血胶囊可明显降低血浆Hcy水平,抑制Hcy诱导的AS形成,其机制可能与下调VCAM-1表达有关。
Objective To explore the effect of tonifying kidney activating blood circulation capsule on aortic vascular cell adhesion molecule (VCAM)-1 mRNA and protein expression in atherogenesis of homocysteine (Hcy)-induced atherosclerosis rabbits. Methods Forty male New Zealand white rabbits were divided into normal control, model, low and high dosage of tonifying kidney activating blood circulation capsule and western medicine (folic acid, Vit B6 and Vit B12) groups. Apart from normal control group, the rabbits in the other groups were induced hyperhomocysteinemia and atherosclerosis model followed by treatment with different drugs. Plasma Hcy concentration, aortic morphological changes, VCAM-1 mRNA and protein expression in abdominal aorta were examined by high performance liquid chromatography, HE staining, real-time quantitative PCR and Western blot, respectively. Results Both tonifying kidney activating blood circulation capsule and western medicine significantly decreased the plasma Hcy concentration, attenuated the intimal thickening, down-regulated VCAM-1 mRNA and protein expression. The efficacy of high dose of tonifying kidney activating blood circulation capsule was superior to that of low dose and western medicine groups. Conclusions Tonifying kidney activating blood circulation capsule could decrease plasma Hcy level and subsequently reverse the aortic lesion induced by Hey. The mechanisms may be related to its suppressive effect on VCAM-1 expression.
