目的探讨细胞周期蛋白D1(Cyclin D1,CCND1)在U87胶质瘤细胞周期调控中的作用。方法根据NCBI GenBank中cyclin D1序列设计双链siRNA,利用Western blotting技术验证设计的siRNA序列的有效性,并通过流式细胞术检测抑制CCND1对U87胶质瘤细胞周期进程的影响。结果经Western blotting检测,成功抑制了CCND1的表达。并且抑制CCND1后,U87细胞周期停滞在G1期,抑制了U87细胞的G1/S期转换。结论U87细胞中抑制CCND1可以显著抑制细胞周期进程,使细胞周期停滞在G0/G1期,为进一步研究胶质瘤的靶向治疗奠定了理论基础。
Objective To investigate the cell cycle regulatory mechanism of cyclin D1 in U87 glioma cell. Methods We were designed the siRNA of cyclin D1 according to the cDNA sequence of CCND1 from the gene database of NCBI GenBank, and verified the inhibitory effeciency of siRNA using western blotting. Then cell cycle analysis of the CCND1 knockdown U87 cells was carried out by flow cytometry. Results The expression of CCND1 was inhibited, and compared with the negative control (NC) groups transfected with CCND1 siRNA in U87 ceils can significantly arrested the cell cycle at the C1/S transition, and promoted the accumulation of cells in the G0/G1 peak. Conclusion Inhibition of the expression of CCND1 can significantly repressed the cell cycle progression, and will be of special help for treatment of the glioma in clinic.
