目的:研究不同浓度内皮素-1(endothelin-1,ET-1)对牙周膜细胞(periodontal ligament cells,PDLCs)碱磷酶(alk aline ph osph atase,ALP)活性和骨钙素(osteocalcin,OCN)含量的影响。方法:体外培养牙周膜细胞,加入不同浓度(1,10,100n M)的ET-1,以未作任何处理的牙周膜细胞为对照组,刺激48h后观察ET-1对牙周膜细胞ALP活性和OCN含量的影响。结果:经ET-1(1,10,100n M)干预后,PDL细胞ALP活性和OCN含量低于对照组(P〈0.05),并呈剂量依赖性。结论:ET-1对牙周膜细胞ALP活性和OCN含量具有抑制作用。
Objective: To investigate how endothelin-1 affects the alkaline phosphatase( ALP) activity and the production of osteocalcin(OCN) of periodontal ligament(PDL) cells. Methods: The PDL cells were stimulated with endothelin-1(1, 10, 100 n M) for 48 hours. The untreated h PDL cells served as normal control. ALP activity was analysed by colorimetric assay using ALP kit and the amount of OCN was assessed by osteocalcin ELISA kit. Results: The ALP activity and the production of OCN of PDL cells were reduced by ET-1 stimulation in dose-dependent manner. Conclusion: These results indicated that ET-1 could reduce the ALP activity and the production of OCN of PDL cells.
