目的观察密蒙花总黄酮对去势雄鼠干眼症大鼠模型基础泪液分泌量、泪膜稳定性及血清黄体生成素水平的影响。方法将1月龄健康雄性Wistar大鼠150只,随机分为正常组(A组)、假手术组(B组)、手术对照组(C组)、雄激素治疗组(D组)、密蒙花总黄酮治疗组(E组);C、D、E组采用去势的方法建立动物模型,各组在造模后1周开始用药。各组实验动物分别于用药1、3、5个月后随机取10只,行Schirmer I试验、测量泪膜破裂时间,于第5个月,抽血测定血清黄体生成素水平。结果 D、E组Schirmer I试验测量值明显高于C组(P〈0.05)、泪膜破裂时间明显长于C组(P〈0.05)、血清黄体生成素水平明显低于C组(P〈0.0l)。结论雄激素水平下降可导致干眼病,采用双侧睾丸及附睾切除的方法可成功建立雄激素水平下降所致的大鼠干眼病动物模型;密蒙花总黄酮具有拟雄激素效应,能够产生同丙酸睾酮治疗干眼病相似的效果。
Objective To observe the effects of total Buddleja flavonoids on the amount of basal tear secretion,tear film stability,level of luteinizing hormone of castrated male rat modeled with xeroma.Methods 150 Wistar male one-month-old rats were randomly divided into 5 groups including normal group(A),sham operation group(B),surgery control group(C),positive group treated with androgen(D) and Buddleja flavonoids group(E) with 30 rats in each group.The C,D,E groups were cut the bilateral testicle and epididymis,and B group was only incised scrotal without removal of the testicles.A group was not done with anything.One week after modeling whenthe wound was to be healed,all rats began to be given mediations.At the 1st,3rd and 5th months after treatment,10 rats were randomly selected from each group and performed Schirmer I test and tear breakup time measurement.Luteinizing hormones of blood serum were tested in the fifth month.Results The values of Schirmer I tests in D and E groups were significantly higher than that of C group(P〈0.05),tear film breakup time was significantly longer than that of C group(P〈0.05),and levels of luteinizing hormones were less than that of C group(P0.0l).Conclusion Decreased androgen levels can lead to xeroma,and the animal models of xeroma in rats caused by decreased androgen levels can be successfully established with the method of removal of bilateral tests and epididymis.Total flavonoids of Buddleja have androgenic effects,which produces the similar treatment effects on xeroma with testosterone propionate.
