目的探讨脑淋巴引流阻滞(CLB)对蛛网膜下腔出血(SAH)后神经细胞凋亡的影响和吡哆醇的缓解作用及相关机制。方法健康成年Wistar大鼠,随机分为正常对照组、SAH组、SAH+CLB组、SAH+CLB+吡哆醇组、SAH+CLB+生理盐水组。采用枕大池2次注血法建立SAH模型。于第2次注血3 d后检测相关指标。SAH+CLB+吡哆醇组于CLB术前30 min按50 mg.kg-1腹腔注射吡哆醇生理盐水溶液,每天以上述半量重复注射2次。采用TUNEL荧光标记法检测原位凋亡情况;免疫组织化学激光共聚焦检测大鼠皮层神经细胞Caspase-3和Bcl-2的蛋白表达。结果①与正常对照组比较,其余各组大鼠TUNEL阳性细胞表达均明显升高,而SAH+CLB组和SAH+CLB+生理盐水组又明显高于SAH组(P〈0.01),SAH+CLB+吡哆醇组与SAH+CLB组比较表达有所减弱(P〈0.01)。②与正常对照组比较,其余各组大鼠皮层神经细胞Caspase-3蛋白表达均明显升高,而SAH+CLB组和SAH+CLB+生理盐水组最高,与SAH组比较差异具有显著性(P〈0.01),SAH+CLB+吡哆醇组与SAH+CLB组比较表达有所减弱(P〈0.01)。③与正常对照组比较,其余各组大鼠皮层神经细胞Bcl-2蛋白表达均明显升高,而SAH+CLB组和SAH+CLB+生理盐水组明显低于SAH组(P〈0.01),SAH+CLB+吡哆醇组较SAH+CLB组表达增强。结论脑淋巴引流阻滞可以通过Caspase-3高表达和Bcl-2低表达加重SAH后大鼠皮层神经细胞的凋亡,而吡哆醇具有一定的缓解作用。
Aim To investigate the influence of cerebral lymphatic blockade(CLB)on apoptosis of cortical neurons after subarachnoid hemorrhage(SAH)in rats in vivo,and the mitigation role of pyridoxine in rats.Methods Healthy adult Wistar rats were randomly divided into normal control group,SAH group,SAH+CLB group,SAH+CLB+pyridoxine group and SAH+CLB+physiological saline group.SAH model was made by double injection of autologous blood into the cisterna magna.On the third day after the second injection,terminal deoxynucleotidy transferase mediated nick end labeling(TUNEL)method fluorescent was applied to determine in situ apoptosis in the cerebral cortex.Immunohistochemistry was conducted to detect the expression of Caspase-3 and Bcl-2 in cortical neurons.Results The numbers in TUNEL positive cells were less in normal control group than those in remaining group,while the numbers in SAH+CLB group were signifi cantly higher than those in SAH group.The numbers in SAH+CLB+pyridoxine group were less than those in SAH+CLB group.Caspase-3 expression in normal control group was weaker than that in remaining groups,while expression in SAH+CLB group was significantly higher than that in SAH group.Caspase-3 expression in SAH+CLB+pyridoxine group was lower than that in SAH+CLB group.Bcl-2 expression in SAH+CLB group was significantly lower than that in the SAH group,while the expression in SAH+CLB+pyridoxine group was higher than that in SAH+CLB group.Conclusion CLB exacerbates the apoptosis of cortical neurons in rats after SAH by up-regulating Caspase-3 expression and down-regulating Bcl-2 expression,while the pyridoxine has a certain role in the mitigation.
