[目的]观察铁对体外培养人外周血淋巴细胞增殖的影响,初步探讨铁对人体免疫功能影响的机制。[方法]用去铁胺去除RPMI1640培养基中的铁,通过细胞增殖实验(MTT)和流式细胞术的方法来观察不同浓度铁对人外周血淋巴细胞增殖的影响。[结果]细胞增殖实验结果显示,C组(生理浓度组)淋巴细胞增殖最好,5组呈倒“V”字形趋势(P〈0.05)。铁刺激T细胞增殖时A组、D组和E组与C组之闻差异和刺激B细胞增殖时A组、B组、D组和E组与C组之间差异均有统计学意义(P〈0.05)。流式细胞术结果显示,5组间CD4+T细胞比例呈倒“V”字型剂量反.应关系,以C组比例最高(P〈0.05)。随铁浓度增大,5组间CD8+T细胞比例线性升高,而CD4+/CD8+线性降低。[结论]生理浓度的铁最适于人外周血淋巴细胞增殖,而铁缺乏和铁过重均不利于人外周血淋巴细胞增殖。
[ Objective] To study the effect of iron on the proliferation of peripheral blood lymphocyte in vitro, and preliminarily explore the mechanism of iron influencing human immune function. [ Methods] Deferoxamine was used to remove the iron in RPMI1640 medium, The effect of different doses of iron on the proliferation of peripheral blood lymphocyte in vitro were detected by using MTT and flow cytometry. [Results] The results of proliferation of cell showed that the group C (physiological concentration had the best effect of lymphocyte proliferation, an inverse "V" tendency was found among 5 groups (P 〈 0.05). Significant difference was existed in T cells between group A, D, E and group C (P 〈 0.05) when stimulated by PHA, also, significant difference was revealed in B cells between group A, B, D, E and group C (P 〈 0.05) when stimulated by LPS. Through flow eytometry an inverse "V" tendency was also found in CD4+ T cells between 5 groups (P〈 0.05) in which group C accounted for the highest percentage, and percentage of CD8+ T cells raised while proportion of CD4+/CD8+ declined linearly with the increasing of iron concentration. [Conclusion] This study indicates that proliferation of peripheral blood lymphocyte performed best in physiological concentration while iron deficiency and iron overdose have disadvantage in the proliferation of peripheral blood lymphocyte in vitro.
