目的探讨miR.10家族中的miR-10a、miR.10b在人垂体腺瘤中的表达情况,并分析其与垂体腺瘤侵袭性的关系。方法收集经病理确诊的21例垂体腺瘤及3例来自于尸体解剖的正常腺垂体组织,运用实时荧光定量PCR技术检测miR—lOa、miR-10b在人垂体腺瘤及正常腺垂体组织中的表达,并进-步分析miR-10家族在不同分类垂体腺瘤中的表达差异。结果miR-10b在垂体腺瘤中较正常腺垂体表达明显增高,且在巨大垂体腺瘤中的表达量明显高于微腺瘤和大腺瘤;miR-10a、miR-10b在侵袭性垂体腺瘤中表达水平均明显高于非侵袭组和正常组,且与肿瘤侵袭程度呈正相关。结论miR.10a,miR-10b在侵袭性垂体腺瘤组织中表达增高,二者均与垂体腺瘤的侵袭程度呈正相关;特别是miR-10b,在侵袭性垂体腺瘤中表达升高更加明显并且与肿瘤直径呈正相关。
Objective To investigate the expression of miR-10a and miR-10b in specimens of pituitary ade- noma, analysis the relationship between miR-10 family expressing and pituitary adenoma invasion potential. Methods 21 eases of pituitary adenoma samples which were confirmed by pathological results and 3 samples of normal pituitary tissues which were collected after neurosurgical operation were enrolled in this study. Total miRNAs were extracted using one step miRNA out method, the relative expression of miR-lOa, miR-lOb were quantified by real-time fluorescence quantitative RT-PCR, and the correlation of miR-10 family members' ex- pression with invasion and diameter of pituitary adenoma were analyzed by SPSS software. Results miR-10b was up-regulated in pituitary adenoma and over-expressed in macroadenoma; miR-10a and miR-10b were both over-expressed in invasion group. It showed an upward trend as pituitary adenoma invasive grand increased. Conclusions miR-10b is over-expressed in pituitary adenoma and has a positive linearity relationship with tumor diameter; miR-10a and miR-10b are both correlated with invasion grade of pituitary adenoma.
