Klotho gene is highly expressed in the brain and kid-ney, with the productions of membrane-bound Klotho and solu-ble Klotho. Membrane-bound Klotho acts as a receptor, while soluble Klotho functions as a humoral factor. Klotho proteins modulate a variety of diseases via regulating calcium and phos-phorus metabolism and cardiovascular protection. Recent studiesindicate the association between Klotho and diseases such as car-diovascular diseases, cancer, kidney damage, and metabolic diseases. The role of Klotho protein in these diseases will be re-viewed in this article.
