利用活体成像技术检测姜黄素药物组织分布的方法探索

中文摘要:

目的探讨应用目前比较前沿的小动物活体成像技术进行姜黄素药物组织分布检测的方法探索,建立实验方案,评价其应用性。方法采用CY7荧光染料进行姜黄素标记,并进行纯化,利用高效液相进行测定,裸鼠注射后进行荧光活体成像检测,对其药物分布情况进行实时摄取图像,并选取心、肝脏、脾脏、肺、肾脏等进行解剖后荧光成像,然后对其冰冻切片,进行共聚焦显微镜观察。结果 CY7标记成功,纯度达90%以上,体内活体成像,各器官、膀胱呈现比较好的浓度代谢规律,切片后能够对药物进入细胞情况进行比较显著的观测。结论该实验方案切实可行,方便经济,对于中药的药代动力学检测具有重要应用价值。

英文摘要:

OBJECTIVE To explore the method of determining curcumin pharmacokinetics by more advanced in-vivo imaging of small animals at present to establish experimental schema and evaluate its application.METHODS CY7 fluorescent dyes were applied to label and purify curcumin,and high-performance liquid chromatography(HPLC) was used to determine.After naked mice were injected,fluorescent in-vivo imaging detection was performed to get their real-time images on the distribution of drugs,and fluorescence imaging was made after hearts,livers,spleens,lungs as well as kidneys were dissected,then slices were froze to carry on confocal microscopy observation.RESULTS The label of CY7 was successful,whose purity was more than 90%.Each organ and the bladder can present better concentration metabolic regularity after in-vivo imaging in the body.The condition of drugs into slices can be significantly observed after cutting into slices.CONCLUSION This experimental scheme is feasible,convenient and economical,which has important application value for the pharmacokinetic detection of Chinese drugs.
